空育131和稻花香是黑龙江省很有代表性的寒地栽培品种,也是育种研究的重要材料。为了获取此品种的优质RNA,以经低温避光处理3d的空育131及稻花香水稻的10日龄水稻苗叶为材料,利用优化的UNIQ-10柱式Total RNA Extractor(Trizol)试剂盒法提取RNA。之后,进行RT-PCR反应,并对RT-PCR的结果进行琼脂糖凝胶电泳的检测。结果表明:通过优化,改进了UNIQ-10柱式Total RNA Extractor(Trizol)试剂盒法,提取的RNA纯度较高,A260/A280均接近1.8,并且RT-PCR的条带清晰。
Kongyu 131 and Daohua are the most representative winter cultivars in Heilongjiang Province and are also important materials for breeding research. In order to obtain high-quality RNA of this variety, the 10-day-old rice seedlings of Kong-yu-131 and Dangao-scented rice treated with low temperature and darkness for 3 days were extracted with optimized UNIQ-10 Total RNA Extractor (Trizol) RNA. Thereafter, the RT-PCR reaction was performed and the result of the RT-PCR was subjected to agarose gel electrophoresis. The results showed that the total RNA extracted by UNIQ-10 Total RNA Extractor (Trizol) kit was improved by using the optimized method. The purity of the extracted RNA was high, A260 / A280 were close to 1.8, and the band of RT-PCR was clear.
