目的 分析羊水过少产妇胎盘、胎膜组织中Caspase7、Cx43蛋白和mRNA的表达特点,揭示羊水过少的发生机制,为中晚期羊水过少产妇的治疗提供理论依据.方法 选取2016年1月-2017年1月在该院产科分娩的足月产妇120例为研究对象,其中羊水过少产妇49例(过少组),羊水正常产妇71例(正常组).采用免疫组化染色法和RT-PCR法检测两组产妇胎盘、胎膜组织中Caspase7、Cx43蛋白及mRNA表达水平,并探讨其临床意义.结果 过少组胎膜及胎盘组织中的Cx43蛋白阳性表达率分别为10.20%、12.24%,显著低于正常组的26.76%、30.99%,差异具有统计学意义(均P<0.05);过少组胎膜及胎盘组织中的Cx43 mRNA表达水平显著低于正常组(t=10.103、9.478,均P<0.05),过少组胎膜及胎盘组织中的Caspase7 mRNA表达水平显著高于正常组(t=12.378、10.736,均P<0.05);过少组胎膜及胎盘组织中的Caspase7蛋白阳性表达率分别为53.06%、57.14%,均显著高于正常组的23.94%、25.35%,差异具有统计学意义(均P<0.05).结论 羊水过少产妇胎盘、胎膜组织中Caspase7蛋白及mRNA高表达、Cx43蛋白及mRNA低表达,相关指标的表达差异可能与羊水过少的发生具有一定的相关性.“,”Objective To analyze the expression characteristics of Caspase7,Cx43 proteins and mRNA in placental and fetal membrane tissues of postpartum women with oligohydramnios,reveal the mechanism,and provide a theoretical basis for treatment of oligohydramnios during the second and the third trimesters of pregnancy.Methods A total of 120 postpartum women of full-term pregnancy gave birth to their babies in Department of Obstetrics in Beijing Jishuitan Hospital from January 2016 to January 2017,then they were divided into oligohydramnios group (49 cases with oligohydramnios) and normal group (71 cases with normal amniotic fluid volume).Immunohistoclhemical staining and RT-PCR were used to detect the expression levels of Caspase7,Cx43 proteins and mRNA in placental and fetal membrane tissues in the two groups.The clinical significance was discussed.Results The positive expression rates of Cx43 protein in placental and fetal membrane tissues in oligohydramnios group were 10.20% and 12.24%,respectively,which were statistically significantly lower than those in normal group (26.76% and 30.99%,respectively) (both P<0.05).The expression levels of Cx43 mRNA in placental and fetal membrane tissues in oligohydramnios group were statistically significantly lower than those in normal group (t =10.103,9.478,both P<0.05).The expression levels of Caspase7 mRNA in placental and fetal membrane tissues in oligohydramnios group were statistically significantly higher than those in normal group (t =12.378,10.736,both P<0.05).The positive expression rates of Caspase7 protein in placental and fetal membrane tissues in oligohydramnios group were 53.06% and 57.14%,respectively,which were statistically significantly higher than those in normal group (23.94% and 25.35%,respectively) (both P<0.05).Conclusion The high expression of Caspase7 protein and mRNA,low expression of Cx43 protein and mRNA in placental and fetal membrane tissues of postpartum women may be related to oligohydramnios to a certain extent.
