目的 :观察HPV 16E6和食管癌细胞核基质的关系 ,以探讨HPV感染在食管癌变中的作用。 方法 :2株食管癌细胞系 ,EC/CUHK1和EC/CUHK2 ,用于本实验。PCR扩增核基质相关的HPV 16E6亚基因片段 ,RT PCR和免疫细胞化学染色观察了E6亚基因在癌细胞内的表达。结果 :EC/CUHK2细胞的HPV 16E6亚基因片段存在于核基质相关DNA中 ,E6癌蛋白位于核基质组分内 ;而EC/CUHK1细胞的结果均为阴性。结论 :HPV 16E6和核基质的相互作用有助于病毒导致的食管癌发生。
Objective: To observe the relationship between HPV 16E6 and the nuclear matrix of esophageal cancer cells in order to investigate the role of HPV infection in esophageal carcinogenesis. Methods: Two esophageal cancer cell lines, EC / CUHK1 and EC / CUHK2, were used in this experiment. PCR amplification of nuclear matrix-associated HPV 16E6 subunit fragments, RT PCR and immunocytochemical staining E6 subunit expression in cancer cells. Results: The HPV 16E6 subunit fragment of EC / CUHK2 cells was present in nuclear matrix-associated DNA and the E6 oncoprotein was located in the nuclear matrix components. The results of EC / CUHK1 cells were negative. Conclusion: The interaction of HPV 16E6 and nuclear matrix contributes to the virus-induced esophageal carcinogenesis.
