目的对比PFGE和MLVA两种不同分型方法用于大理地区合并HIV感染肺结核患者Mtb临床分离株基因分型的优劣。方法选取大理地区27株合并HIV感染肺结核患者Mtb临床分离株,采用PFGE和MLVA两种方法进行基因分型,应用BioNumerics(6.6)软件进行聚类分析。结果 PFGE法将27株Mtb分为5个基因群,其中主要优势基因群为A群(15株)包括H37Rv,D=0.698;MLVA法将27株Mtb分为4个基因群,其中主要优势基因群为Ⅰ群(8株,包括H37Rv减毒株)和Ⅱ群(9株)。D=0.999;对比分析显示MLVA法分辨力高于PFGE法。结论 MLVA法与PFGE法相比更适合于大理地区合并HIV感染肺结核患者Mtb的基因分型。
Objective To compare the genotypes of Mtb clinical isolates from patients with HIV-infected pulmonary tuberculosis in Dali with two different genotyping methods, PFGE and MLVA. METHODS: Twenty-seven clinical isolates of Mtb from patients with HIV-infected pulmonary tuberculosis were enrolled in this study. PFGE and MLVA were used for genotyping. BioNumerics (6.6) software was used for cluster analysis. Results The 27 Mtb strains were divided into 5 groups according to the PFGE method. Among them, A group (15 strains) including H37Rv and D = 0.698 were dominant group, and 27 strains were divided into 4 groups by MLVA method. Among them, the dominant gene Groups were Group I (8 strains including attenuated strain H37Rv) and Group II (9 strains). D = 0.999; comparative analysis showed that MLVA method higher resolution than the PFGE method. Conclusion Compared with PFGE, MLVA is more suitable for genotyping Mtb in patients with HIV-infected pulmonary tuberculosis in Dali.
