目的研究蓝靛果花色苷诱导人结肠癌细胞HT29凋亡的形态学特征。方法采用单四唑(MTT)法检测蓝靛果花色苷对HT29细胞生长的抑制作用;采用倒置显微镜、荧光倒置显微镜、单细胞电泳、扫描电镜、透射电镜观察不同浓度(0.5,1.0和2.0mg/ml)蓝靛果花色苷处理HT29细胞凋亡形态学特征的变化。结果蓝靛果花色苷可抑制HT29细胞生长,抑制率呈时间剂量依赖性。蓝靛果花色苷作用于结肠癌HT29细胞,使结肠癌细胞分别出现了细胞数量明显减少,体积缩小,皱缩变形,细胞表面有许多的球状突起,进一步有凋亡小体产生,细胞间连接逐渐断裂,细胞浆出现较多空泡,胞浆浓缩,核物质致密,呈斑块状,或呈新月形边集于核膜,核形不规整等典型的凋亡细胞形态特征。随着花色苷浓度的增加,现象更加显著。结论蓝靛果花色苷可诱导结肠癌HT29细胞凋亡,显著抑制结肠癌HT29细胞的增殖。
Objective To investigate the morphological characteristics of apoptosis induced by anthocyanin in Lonicera edulis HT29 cells. Methods MTT assay was used to detect the inhibitory effect of anthocyanins on the growth of HT29 cells. The effects of different concentrations (0.5, 1.0 and 2.0 mg / kg) on the growth of HT29 cells were observed by inverted microscope, fluorescence inverted microscope, single cell electrophoresis, scanning electron microscopy and transmission electron microscopy. ml) treatment of apoptosis in HT29 cells with morphological features. The cytoplasm showed more vacuoles, cytoplasm condensation, dense nuclear material, patchy or crescent-shaped nuclei, irregular nuclei and other typical morphological features of apoptotic cells. With the increase of anthocyanin concentration, the phenomenon is more significant. Conclusion Anthocyanin of Lonicera edulis can induce apoptosis of colon cancer HT29 cells and significantly inhibit the proliferation of colon cancer HT29 cells.
