以无刺桅杆槐和无刺槐的带腋芽茎段为材料获取无菌苗,再以无菌苗叶片为外植体进行植株再生研究。结果表明:2种无刺槐茎段的最佳取材时间均为5月上旬;无刺桅杆槐适宜的叶片愈伤组织诱导培养基为MS+6-BA 0.25 mg/L+2,4-D 6.00 mg/L,无刺槐为MS+6-BA 0.50 mg/L+2,4-D 2.00 mg/L,愈伤诱导率分别为60%、80%;不定芽诱导培养基,无刺桅杆槐和无刺槐均为MS+6-BA 5.00 mg/L+IBA 0.10 mg/L+GA30.10 mg/L,再生率分别为60%、50%。2种无刺槐的生根培养基均为1/2MS+IBA 0.05 mg/L,生根率分别为55.6%、72.2%,组培苗移栽成活率达85%以上。
Aseptic seedlings were obtained from the axillary buds of Acanthopanax setae and Acacia crassicarpa and were used as explants to study the plant regeneration. The results showed that the optimum time for the extraction of stems of two locust-free locusts was in the first ten days of May. The suitable callus induction mediums for the non-stalked locusts were MS + 6-BA 0.25 mg / L + 2,4-D 6.00 The callus induction rate was 60% and 80% for MSA-6-BA 0.50 mg / L + 2,4-D 2.00 mg / L without R. pseudoacacia. Adventitious bud induction medium, All the locust-free locusts were MS + 6-BA 5.00 mg / L + IBA 0.10 mg / L + GA 30.10 mg / L, with regeneration rates of 60% and 50%, respectively. The rooting rate of two kinds of non-acacia locusts were 1 / 2MS + IBA 0.05 mg / L, the rooting rates were 55.6% and 72.2% respectively, and the transplanting survival rate of tissue culture seedlings was more than 85%.
