目的研究葛根素对血管性痴呆(VD)大鼠行为学及额叶细胞凋亡的影响。方法采用大鼠双侧颈总动脉(CCA)永久性阻断法(2-VO)建立VD大鼠模型,大鼠随机分为假手术组、2-2VO组和葛根素组,各组大鼠又分为4、8周2个时间点。用暗回避反应法测定学习记忆能力,并应用免疫组织化学法、原位末端标记法检测大鼠额叶bcl-2蛋白表达水平及凋亡细胞数。结果2-2VO组和葛根素组大鼠潜伏期明显缩短,但与2-2VO组比较葛根素大鼠潜伏期均显著延长。2-2VO组和葛根素组大鼠额叶bcl-2的表达水平及凋亡细胞数均较假手术组明显增高,葛根素组bcl-2的表达水平较2-2VO组明显增高,而凋亡细胞数较2-2VO组明显减少。结论葛根素可通过上调bcl-2基因表达而抑制细胞凋亡,改善VD大鼠的学习记忆能力。
Objective To study the effect of puerarin on behavior and apoptosis of frontal lobe in vascular dementia (VD) rats. Methods The rat model of VD was established by permanent occlusion of bilateral common carotid artery (CCA). Rats were randomly divided into sham operation group, 2-2VO group and puerarin group. Rats in each group It is divided into 4 and 8 weeks and 2 time points. The ability of learning and memory was measured by the dark avoidance reaction method. The expression of bcl-2 protein and the number of apoptotic cells in the frontal lobe were detected by immunohistochemistry and in situ end labeling. Results The latency period of 2-2VO group and puerarin group was significantly shortened, but the latencies of puerarin rats were significantly longer compared with 2-2VO group. The expression of bcl-2 in the frontal lobe and the number of apoptotic cells in the 2-2VO group and the puerarin group were significantly higher than those in the sham operation group. The expression of bcl-2 in the puerarin group was significantly higher than that in the 2-2VO group. The number of dead cells was significantly reduced compared with the 2-2VO group. Conclusion Puerarin can inhibit cell apoptosis by up-regulating bcl-2 gene expression and improve learning and memory ability of VD rats.
