目的:观察桃红四物汤对兔自体皮片移植术后移植皮片的血管密度、血清中MMP-2含量的影响,探讨桃红四物汤对皮片移植术后血管新生的影响机制。方法:将健康新西兰兔75只,按随机数字表法分为5组:桃红四物汤低、中、高剂量组、模型组、假手术组,每组15只。假手术组不进行皮片移植,切开后直接缝合,术后予蒸馏水灌胃,桃红四物汤低、中、高剂量组造模后予以相应剂量的桃红四物汤灌胃,模型组造模后以蒸馏水灌胃,各组灌胃时间最长均为8 d。分别于造模后2 d、5 d、8 d从每组随机抽取5只,检测移植皮片血管密度及血清中MMP-2含量。结果:造模后2 d、5 d、8 d,桃红四物汤低、中、高剂量组血管密度、血清MMP-2浓度均高于模型组(P<0.05);造模后2 d桃红四物汤低、中、高剂量组与模型组血管密度均低于假手术组(P<0.05);造模后2 d、5 d桃红四物汤中、高剂量组血管密度均高于桃红四物汤低剂量组(P<0.05)。造模后2 d、5 d、8 d桃红四物汤低、中、高剂量组血清MMP-2浓度均高于假手术组(P<0.05);造模后2 d、5 d、8 d桃红四物汤中、高剂量组血清中MMP-2浓度均高于桃红四物汤低剂量组(P<0.05);桃红四物汤高、中剂量组血管密度、血清MMP-2浓度比较,差异均无统计学意义(P>0.05)。结论:桃红四物汤能上调自体皮片移植模型兔血清中MMP-2含量,并能使血管密度值增加,具有促血管新生作用。
Objective: To observe the influence of Taohong Siwu Decoction on vascular density and serum MMP-2 content in autologous skin graft after autologous skin grafting to explore the mechanism of Taohong Siwu Decoction on angiogenesis after skin grafting. Methods: A total of 75 healthy New Zealand rabbits were randomly divided into 5 groups according to the random number table: low, medium and high dose of Taohong Siwu Decoction, model group and sham operation group. Sham-operation group was not skin grafting, sutured directly after incision, and distilled water was administrated after operation. The rats in low, medium and high dose group of Taohong Siwu decoction were given gavage with Taohong Siwu Tang, The model was administered with distilled water, the longest time of each group was 8 d. Five rabbits were randomly selected from each group on the 2nd, 5th, and 8th day after model establishment, and the vascular density and the level of MMP-2 in the grafted skin were measured. Results: The vascular density and serum MMP-2 concentration in the low, medium and high dose Taohongsiwu Decoction group were significantly higher than those in the model group (P <0.05) at 2 days, 5 days and 8 days after modeling. The vessel densities of Siwu Tang low, medium and high dose groups and model group were lower than those of sham operation group (P <0.05). The vascular densities of middle and high dose Taohongsiwutang groups were higher than those of Taohong Siwu Tang low-dose group (P <0.05). The levels of serum MMP-2 in the low, middle and high doses of Taohong Siwu decoction at 2 days, 5 days and 8 days after modeling were higher than those in the sham operation group (P <0.05). After 2 days, 5 days and 8 days The concentrations of MMP-2 in serum of Taohong Siwu Decoction group were higher than that of Taohong Siwu Decoction low dose group (P <0.05) There was no significant difference (P> 0.05). Conclusion: Taohong Siwu Tang can up-regulate the content of MMP-2 in sera of rabbits with autologous skin grafts, and can increase the value of blood vessel density and promote angiogenesis.
