目的 观察内毒素血症大鼠 (endotoxemia)的血液流变性变化。方法 给大鼠一次性静注内毒素 (LPS ,0 .5mg kg ,iv) ,分为LPS(lipopolysaccharide ,LPS) +多粘菌素B组、LPS +生理盐水组、LPS模型组 ,给大鼠一次性静注内毒素 (lipopolysaccharide ,LPS ,0 .5mg kg,iv) ,前 2组分别于注入内毒素后 0 .5h静脉给予多粘菌素B(polymyxinBsulfate ,0 .2mg kg)和 1ml生理盐水 (salinesolution ,NS) ,LPS模型组大鼠于注入内毒素后不作其它处理。在给内毒素后 1、2、4h抽血液 ,并检测全血粘度 (wholebloodviscosity ,WBV) ,聚集指数 (aggregationindexofredcells,AI)和红细胞压积 (hematocritofredcells,HRC)。结果 LPS +多粘菌素B组 ,可明显降低内毒素血症后 1h的全血粘度 (高切变、低切变 )、红细胞压积、聚集指数显著升高 ,MAP于 2、4h明显高于LPS +生理盐水组和空白对照组。结论 多粘菌素B治疗能明显改善内毒素血症早期的血液流变性 ,增强红细胞变形能力 ,降低聚集性 ,改善微循环 ,对预防内毒素血症有一定作用。
Objective To observe the changes of hemorheology in endotoxemia rats. Methods LPS (0.5 mg kg, iv) was intravenously injected intravenously into rats and divided into LPS group, LPS + saline group and LPS group. One time of intravenous injection of lipopolysaccharide (LPS, 0.5 mg kg, iv), the first two groups were given intravenous injection of polymyxin Bsulfate (0.2 mg kg) and 1 ml normal saline (salinesolution, NS). Rats in LPS model group were not treated with endotoxin. Blood was drawn at 1, 2 and 4 h after endotoxin administration, and whole blood viscosity (WBV), aggregation index (AI) and hematocrit of red cells (HRC) were measured. Results In the LPS + polymyxin B group, the whole blood viscosity (high shear and low shear) at 1 hour after endotoxemia was significantly decreased, the hematocrit and aggregation index were significantly increased, and MAP was significantly higher at 2 and 4 hours LPS + saline group and blank control group. Conclusion Polymyxin B treatment can significantly improve the early blood rheology of endotoxemia, enhance the ability of erythrocyte deformability, reduce aggregation, improve microcirculation and prevent endotoxemia to a certain extent.
