目的探讨活动期SLE患者外周血中T、B细胞表面Fas和bcl-2的表达水平及其与T、B细胞凋亡的关系。方法采用流式细胞术,测定活动期SLE患者外周血T、B细胞表面Fas和bcl-2的表达,并同时测定患者血中T、B细胞的凋亡。结果①活动期SLE患者外周血中B细胞及CD8+T细胞表面bcl-2的表达明显高于正常人(分别为P∨0.05和P∨0.01);CD4+及CD8+T细胞表面Fas的表达高于正常人(分别为P∨0.05和P∨0.01);B细胞表面Fas的表达降低(P∨0.01);CD4+T细胞bcl-2的表达下降(P∨0.01)。②活动期SLE患者血中B细胞的凋亡率明显下降,CD8+T细胞数较正常对照组增加,CD4+T细胞低于常人(分别为:P∨0.01、P∨0.05和P∨0.01)。结论SLE患者体内淋巴细胞凋亡的异常与T、B细胞表面Fas和bcl-2基因表达的异常有关。
Objective To investigate the expression of Fas and bcl-2 on T and B cells in peripheral blood of patients with active SLE and their relationship with the apoptosis of T and B cells. Methods Flow cytometry was used to detect the expression of Fas and bcl-2 on T and B cells in peripheral blood of patients with active SLE. The apoptosis of T and B cells in the blood of patients with active SLE was also measured. Results ① The expression of bcl-2 on the surface of B cells and CD8 + T cells in peripheral blood of active SLE patients was significantly higher than that of normal controls (P ∨ 0.05 and P ∨ 0.01, respectively). The expression of Fas on the surface of CD4 + and CD8 + T cells was high (P ∨ 0.05 and P ∨ 0.01, respectively); the expression of Fas on B cells decreased (P ∨ 0.01); the expression of bcl-2 decreased in CD4 + T cells (P ∨ 0.01). (2) The apoptotic rate of B cells in active SLE patients decreased significantly, the number of CD8 + T cells increased compared with normal control group, CD4 + T cells were lower than normal people (P ∨ 0.01, P ∨ 0.05 and P ∨ 0.01) . Conclusion The abnormal lymphocyte apoptosis in SLE patients is related to abnormal expression of Fas and bcl-2 gene on T and B cells.
