目的建立鱼、鸭肉和肝脏样品中微囊藻毒素(MC-LR)含量的测定方法。方法取鱼、鸭肉和肝脏经组织捣碎机捣碎,甲醇提取,离心,Sep-pakCl8(ODS)小柱富集、净化,0.1%三氟乙酸(TFA)甲醇梯度解析,酶联免疫吸附试验(ELISA)法测定MC-LR含量。结果方法最小检出浓度为0.1ng/g,相对标准偏差为5.34%,加标回收率为90%~92%。结论该方法预处理样品分离效果好,对测定无干扰,所建立的方法经实际应用取得了满意的效果。
Objective To establish a method for the determination of microcystins (MC-LR) in fish, duck and liver samples. Methods The fish, duck and liver were mashed by tissue masonry, extracted with methanol, centrifuged, and eluted with Sep-pakCl8 (ODS) cartridge. Purification was performed by gradient elution with 0.1% trifluoroacetic acid (TFA) Test (ELISA) determination of MC-LR content. Results The minimum detectable concentration was 0.1 ng / g, the relative standard deviation was 5.34% and the recovery was 90% -92%. Conclusion The method pretreatment sample separation effect is good, no interference on the determination of the established method has been achieved by the practical application of satisfactory results.
