观察人外周血γδT细胞在裸鼠体内的抗肿瘤效应。方法用MEP和IL┐2共刺激培养方法,扩增人外周血γδT细胞,与人肠癌细胞系(LoVo)细胞混合接种于裸鼠体皮下。结果经共刺激培养10天后,细胞总数可增加100~300倍,γδT细胞的比率上升到70%以上。给裸鼠接种1×106LoVo细胞1个月后,不接种γδT细胞的对照组裸鼠皮下瘤块重量为0.4g~0.9g,瘤块病理切片显示有明显肿瘤细胞堆积;而同时接种1×107γδT细胞的实验组裸鼠瘤块重量仅为0~0.005g,瘤块病理切片显示其中除肿瘤细胞外,还伴有增生性淋巴细胞。结论人外周血γδT细胞能有效地抑制人肠癌细胞LoVo在裸鼠体内的生长。
Observe the antitumor effect of human peripheral blood γδT cells in nude mice. Methods Human peripheral blood γδT cells were amplified by MEP and IL┐2 co-stimulation culture method and mixed with human colon cancer cell line (LoVo) cells inoculated subcutaneously in nude mice. Results After 10 days of co-stimulation, the total number of cells increased by 100 to 300 times, and the ratio of γδT cells increased to more than 70%. After nude mice were inoculated with 1×10 6 LoVo cells for 1 month, the nude mice in the control group without γδ T cells inoculated had a subcutaneous tumor weight of 0.4 g to 0.9 g. Tumor blocks showed a clear accumulation of tumor cells; while inoculation 1 The weight of nude mice in the experimental group of ×107γδT cells was only 0-0.005 g. Pathological sections of the tumor showed that in addition to the tumor cells, proliferative lymphocytes were also accompanied. Conclusion Human peripheral blood γδT cells can effectively inhibit the growth of human colon cancer LoVo cells in nude mice.
