目的 :探讨鱼棉平喘方 (DYA)对嗜喘豚鼠的平喘作用及作用机理。方法采用氯化乙酰胆碱和磷酸组胺混合液雾化引喘 ,观察豚鼠引喘潜伏期的变化 ;采用 4 %卵清蛋白生理盐水和 4 %氢氧化铝凝胶致敏 ,卵清蛋白复制哮喘模型 ,生物定量法测定血浆中血小板活化因子 (PAF)的含量。结果 :DYA给药后各组豚鼠的引喘潜伏期比给药前及正常组均显著延长 (P <0 .0 1 )。DYA治疗各组与模型组比较 ,血浆cAMP含量及cAMP/cGMP比值均显著升高 (P <0 .0 1 ) ,血浆cGMP含量显著降低 (P <0 .0 1 )。结论 :DYA有明显的平喘作用 ,其作用机理与其调节机体cAMP、cGMP水平 ,稳定cAMP/cGMP比值 ,降低哮喘模型豚鼠异常升高的血浆PAF水平有关
Objective: To explore the antiasthmatic effects of Yumian Pingchuan Prescription (DYA) on asthmatic guinea pigs and its mechanism of action. Methods The mixture of acetylcholine chloride and histamine was used to atomize and induce asthma. The guinea pigs were observed for the change of asthmatic latency. The sensitization was performed with 4% ovalbumin saline and 4% aluminum hydroxide gel, and ovalbumin was used to replicate the asthma model. Bio-quantitative assay of plasma platelet activating factor (PAF) content. RESULTS: After DYA administration, the asthmatic latency of guinea pigs in each group was significantly longer than that before administration and in the normal group (P < 0.01). Compared with the model group, the plasma cAMP content and cAMP/cGMP ratio in DYA treatment groups were significantly higher (P < 0.01), and plasma cGMP levels were significantly lower (P <0.01). Conclusion: DYA has obvious anti-asthmatic effects, and its mechanism is related to its regulation of cAMP and cGMP levels, stable cAMP/cGMP ratio, and reduction of abnormal elevated plasma PAF levels in guinea pigs with asthma.
