目的:研究汉族健康人能否采用单点法或二点法代替常规采血法计算安替比林药动学参数,并确定其最佳实验方案及采血时间;方法:8例汉族健康自愿者顿服安替比林750mg后,间隔不同时间采血,HPLC法测定其血药浓度,比较一点法或二点法与常规多点采血法计算药动学参数(Cl、t_(1/2)、V_d)的差异;结果:相关分析表明,在选择适当采血时间的条件下,简化法与常规法测定的Cl和t_(1/2)相关性好(r>0.85),误差小(RSD<15%)。采用单点法时,采血时间应大于20h,如24、32h,采用两点法时,可选择4~32、8~32或4~24h。用Dossing公式估算AP的V_d,与常规法相比,相关性差(r=0.663,P>0.05)。结论:汉族健康人可采用简化样本法计算AP的t_(1/2)和Cl,但不宜采用Dossing公式估算AP的V_d。
Objective: To study whether Han healthy people could use single point or two point instead of routine blood sampling to calculate the antipyrine pharmacokinetic parameters and to determine the best experimental plan and blood sampling time. Methods: Eight healthy Han volunteers After taking antipyrine 750mg, blood samples were taken at different time intervals and their plasma concentrations were determined by HPLC. The pharmacokinetic parameters (Cl, t_ (1/2), V_d (R> 0.85). The error was small (RSD <15%), and the difference between Cl and t_ (1/2) ). When using single-point method, blood sampling time should be greater than 20h, such as 24,32h, using two-point method, you can choose 4 ~ 32,8 ~ 32 or 4 ~ 24h. Using Dossing formula to estimate the V_d of AP, compared with the conventional method, the correlation was poor (r = 0.663, P> 0.05). CONCLUSION: Han Chinese healthy subjects can use simplified sample method to calculate t_ (1/2) and Cl of AP, but it is not advisable to estimate V_d of AP using Dossing formula.
