目的研究白细胞介素1α(interleukin -1α ,IL -1α)对椎间盘髓核中蛋白多糖代谢的影响。方法取自然流产的胎儿1具 ,胎龄8个月 ,术后4h内无菌取出椎间盘 ,进行髓核组织块的体外培养 ,在培养液中分别加入IL -1α0.1、0.2、0.4nmol/L ,培养30h后 ,用Alcian法检测培养液中硫酸软骨素的含量。在髓核组织培养液中加入IL -1α0.3nmol/L ,分别培养12、24、72h ,然后检测培养液中硫酸软骨素的含量。结果IL -1α实验组较对照组培养液中硫酸软骨素的含量增加 ,随着IL -1α浓度的增加 ,培养液中硫酸软骨素的含量也增加 ,呈浓度依赖性。随着时间的增加 ,培养液中硫酸软骨素的含量也增加 ,呈现明显的时间依赖性。结论IL -1α可以刺激椎间盘基质中蛋白多糖降解 ,呈现明显的时间和浓度依赖性。
Objective To investigate the effect of interleukin-1α (IL-1α) on proteoglycan metabolism in nucleus pulposus. Methods One fetus with spontaneous abortion and 8 gestational age were randomly selected. The discs were aseptically removed within 4 hours after operation. The in vitro culture of nucleus pulposus was performed. IL-1α0.1, 0.2 and 0.4 nmol / L, cultured for 30h, Alcian was used to detect the content of chondroitin sulfate in the culture medium. IL-1α0.3nmol / L was added to the nucleus pulposus tissue culture medium for 12, 24 and 72h respectively, and then the content of chondroitin sulfate in the culture medium was detected. Results The concentration of chondroitin sulfate in the culture medium of IL-1α group increased compared with the control group. With the increase of IL-1α concentration, the content of chondroitin sulfate in the culture medium also increased in a concentration-dependent manner. With the increase of time, the content of chondroitin sulfate in the culture medium also increased, showing a significant time-dependent. Conclusion IL - 1α can stimulate proteoglycan degradation in the intervertebral disc matrix, showing a significant time and concentration - dependent.
