目的:探讨补中益气汤对裸鼠肺腺癌移植瘤生长抑制作用及对磷脂酰肌醇3激酶(phosphatidylinositol 3 kinase,PI3K)/蛋白激酶B(protein kinase B,AKT)信号通路的影响。方法:50只裸鼠随机分为①正常对照组,②荷瘤对照组,③顺铂组,④顺铂联合补中益气汤组,⑤补中益气汤组。分别给予相应干预后,处死小鼠,计算抑瘤率;应用免疫组化方法、实时定量PCR(real time PCR,RT-PCR)检测移植瘤组织中PI3K,AKT的表达水平。结果:顺铂联合补中益气汤组对移植瘤的抑制率最强,与其他各组比较有显著差异(P<0.05),顺铂和补中益气汤协同作用的q值1.201;移植瘤中PI3K,AKT蛋白和mRNA基因的表达在顺铂联合补中益气汤组显著降低(P<0.05)。结论:补中益气汤联合顺铂对耐顺铂肿瘤有协同抑制作用;补中益气汤能通过干预PI3K、AKT基因和蛋白的表达而增加顺铂对耐顺铂肿瘤的敏感性。
Objective: To investigate the effects of Buzhong Yiqi Decoction on the growth of lung adenocarcinoma xenografts and the effects of phosphatidylinositol 3 kinase (PI3K) / protein kinase B (AKT) signaling pathway in nude mice. Methods: Fifty nude mice were randomly divided into ① normal control group, ② tumor-bearing control group, ③ cisplatin group, ④ cisplatin combined with Buzhongyiqi Decoction group, and Buzhongyiqi Decoction group. The mice were sacrificed and the tumor inhibition rate was calculated after corresponding intervention. The expression of PI3K and AKT in the tumor tissue was detected by real-time PCR (RT-PCR). Results: The inhibitory rate of cisplatin combined with Buzhongyiqi Decoction group was the strongest on the xenografts, which was significantly different from other groups (P <0.05). The q value of synergistic effect of cisplatin and Buzhong Yiqi Decoction was 1.201. The expression of PI3K, AKT protein and mRNA in the tumor group was significantly lower than that in cisplatin combined with Buzhongyiqi Decoction group (P <0.05). Conclusion: Buzhong Yiqi Decoction combined with cisplatin has a synergistic inhibitory effect on cisplatin-resistant tumors. Buzhong Yiqi Decoction can increase the sensitivity of cisplatin to cisplatin-induced tumors by interfering with the expression of PI3K and AKT genes and proteins.
