目的 建立一种简便可行的RP—HPLC法测定肾移植术后病人服用环孢素A(CsA)后全血中的药物浓度 ,为临床合理应用CsA提供一个良好的指标。方法 病人血样经乙醚单步提取后 ,以C8硅胶键合相为固定相 ,乙腈 -甲醇 -水 (40∶34∶2 6 )为流动相 ,6 0℃柱温下进行色谱分析 ,紫外检测波长为 2 14nm。结果 CsA全血浓度在 5 0~ 6 0 0ng·ml-1范围内呈良好的线性关系 ,r =0 .9998,方法的平均回收率为 96 .0 8% ,日内和日间精密度RSD均小于 6 % (n =5 ) ,最低检测限为 10ng·ml-1。结论 本法简便、快速、易于开展 ,用于肾移植术后患者CsA的血药浓度监测 ,效果良好
Objective To establish a simple and feasible method for the determination of the concentration of CsA in whole blood of patients after renal transplantation by RP-HPLC and to provide a good indicator for the clinical application of CsA. Methods The blood samples of patients were extracted by single step with diethyl ether. The mobile phases were acetonitrile-methanol-water (40:34:26), and the chromatographic analysis was performed at 60 ℃. UV detection wavelength For 2 14nm. Results The CsA whole blood concentration showed a good linear relationship in the range of 50-600 ng · ml-1, r = 0.9998. The average recovery was 96.08%. The intra-and inter-day precision RSD Less than 6% (n = 5), the lowest detection limit was 10ng · ml-1. Conclusion This method is simple, rapid and easy to carry out, and is used to monitor the blood concentration of CsA in renal transplant recipients with good effect
