目的 :研究食管癌细胞系SHEEC1以三氧化二砷 (As2 O3 )诱导细胞凋亡光镜和电镜的形态改变。方法 :SHEEC1细胞常规培养在 199培养基 ,在 5 μmoL/LAs2 O3 作用 72h收获细胞以苏木精_伊红染色 ,TUNEL标记 ,透射电镜和流式细胞仪检查。结果 :光镜下凋亡细胞核致密 ,染色质片段化 ,胞浆红染 ,TUNEL标记阳性 ,流式细胞仪检查出现凋亡峰 (亚G1/G0 峰 )。电镜下可见两种死亡细胞 :一种是核固缩 ,有染色质凝集靠近核膜等典型凋亡改变 ;另一种细胞浆退行性变及细胞坏死改变。结论 :食管癌细胞系SHEEC1可以用As2 O3 引起凋亡 ,提示其可以作为治疗食管癌的辅助药物
OBJECTIVE: To study the morphological changes of esophageal cancer cell line SHEEC1 induced by As2O3 under light and electron microscopes. METHODS: SHEEC1 cells were cultured routinely in 199 medium. Cells harvested at 5 μmol / L As2 O3 for 72 h were stained with hematoxylin and eosin, TUNEL, transmission electron microscopy and flow cytometry. Results: Apoptotic nuclei were dense, chromatin fragmentation, cytoplasmic red staining and TUNEL positive staining. Apoptotic peak (sub G1 / G0 peak) was detected by flow cytometry. Under electron microscope, there are two kinds of dead cells: one is nuclear pyknosis, the other is chromatin condensation near the nuclear membrane and other typical apoptotic changes; another cytoplasm degeneration and necrosis changes. Conclusion: The esophageal cancer cell line SHEEC1 can induce apoptosis with As2 O3, suggesting that it may be used as an auxiliary drug for the treatment of esophageal cancer
