目的研究磷酸甘油醛脱氢酶在日本血吸虫感染中的功能。方法荧光定量PCR方法检测磷酸甘油醛脱氢酶在不同来源童虫及日本血吸虫不同发育时期中的转录水平;克隆、表达日本血吸虫磷酸甘油醛脱氢酶,纯化后免疫小鼠,制备免疫血清;采用免疫组织化学法检测磷酸甘油醛脱氢酶在日本血吸虫的组织学定位;于日本血吸虫尾蚴攻击感染前用重组蛋白免疫小鼠,观察其免疫保护效果并探讨其机制。结果磷酸甘油醛脱氢酶在不同来源童虫及日本血吸虫不同发育期差异表达,以宿主组童虫和虫卵表达水平较高。磷酸甘油醛脱氢酶主要位于虫体表膜,可能与其免疫保护性效果相关。重组蛋白免疫小鼠减虫率和减卵率分别为47%和52%。结论磷酸甘油醛脱氢酶在日本血吸虫生长发育中具有重要作用,可能与其生殖功能相关。
Objective To study the function of glyceraldehyde phosphate dehydrogenase in Schistosoma japonicum infection. Methods Fluorescence quantitative PCR was used to detect the transcriptional level of glyceraldehyde phosphate dehydrogenase in Schistosoma japonicum and Schistosoma japonicum at different developmental stages. Clone and expression of glyceraldehyde dehydrogenase of Schistosoma japonicum was purified and immunized to prepare immune serum. Immunohistochemistry was used to detect the histological location of glyceraldehyde-3-phosphate dehydrogenase in Schistosoma japonicum. Before immunization with Schistosoma japonicum, the recombinant protein was used to immunize mice to observe its immunoprotective effect and to explore its mechanism. Results The glyceraldehyde-6-phosphate dehydrogenase was differentially expressed at different developmental stages of Schistosoma japonicum and Schistosoma japonicum, and the expression level of host was higher than that of parasites. Glyceraldehyde-6-phosphate dehydrogenase is mainly located in the epidermis of the parasites and may be related to its immunoprotective effects. The worm reduction rate and the egg reduction rate of recombinant protein immunized mice were 47% and 52% respectively. Conclusion The glyceraldehyde-6-phosphate dehydrogenase plays an important role in the growth and development of Schistosoma japonicum and may be related to its reproductive function.
