CD44 is a well-established cell surface receptor that has been successfully used to design tumor targeting nanocarriers for cancer treatment. However, normal cells such as mesenchymal stem cells and fibroblasts, also demonstrate high expression of the CD44s isoform, leading to potential toxicity of traditional CD44 antibody-based therapy.Specific targeting to the cancer-specific CD44v isoform expressed on tumor cells would substantially reduce toxicity to normal cells. In this study, chondroitin sulfate(CS),a natural polysaccharide, was firstly confirmed to specifically bind to CD44v receptors mostly expressed on tumor cells rather than the standard isoform expressed on normal cells.
