Zuojin Wan (ZJW), a famous traditional Chinese medicine formula, derived from Coptidis rhizoma and Evodiae fructus, has been widely used in treating gastric diseases in China. Previous pharmacological studies identify the alkaloids components correlated with the clinical effect of those herb preparations. In this report a new and sensitive method has been developed for qualitation of 19 components in Zuojin Wan similar formulas(ZJWSFS), including Zuojin Wan(ZJW), Biantong Wan(BTW), Ganlu San(GLS), Zhuyu Wan(ZYW), Fanzuojin Wan(FZJW) by using HPLC in combination with electrospray mass spectrometry (HPLC-ESI-MS), and eight bioactive compounds, I.e., chlorogenic acid (1), jatrorrhizine (2), coptisine (3), palmatin (4), berberine (5), evodin (6), evodiamine (7), rutaecarpine (8) of ZJWSFS and Zuojin Wan preparations(ZJWPS) were determined simultaneously by using HPLC coupled with diode array detector (HPLC- DAD). Simultaneous validation of the 19 components was performed on a C18 column, in which the mobile phase consisted of (A) 0.3% aqueous acetic acid and (B) CH3CN using a gradient elution, the column temperature was set at 20℃, the solvent flow rate was 0.4ml min-1, the ESI-MS spectra were acquired in positive ion mode. A simultaneous determination of the 8 components was performed on a C18 column, in which the mobile phase consisted of (A) 0.1% aqueous phosphoric acid and (B) CH3CN using a gradient elution, the column temperature was set at 20℃, the solvent flow rate was 0.4 ml min-1, detection wavelength was set at 345 nm for chlorogenic acid (1), jatrorrhizine (2), coptisine (3), palmatin (4), berberine (5), and 210 nm for evodin (6), evodiamine (7), rutaecarpine (8), respectively. All calibration curves showed good linear regression (r2 > 0.9970) within test ranges. The method developed showed good precision and accuracy with overall intra- and inter-day variations of 0.54-1.58% and 0.54-2.07%, respectively, and the overall recoveries of 98.05-100.86% for the 8 compounds analyzed. This readily available, low-cost and reliable HPLC-ESI-MS and HPLC-DAD method was useful for authenticity controlling the quality of ZJWSFS and ZJWPS.
