Herbal medicinal products belong to a main part of complementary and alternative medicine and have worldwide using integrated with conventional medicines for treatment. However, the increasing risk of herb-drug interaction, especially mediated by metabolisming enzymes and transporters in pharmacokinetic pathways, has raised enthusiasm on discovering the underlying mechanisms.Natural furanocoumarin compounds are the major bioactive components of Radix Angelicae Dahuricae (RAD), a well-known traditional Chinese medicine existing in many herbal formulas such as Fufang Yangjiao Pian and Jiuwei Qianghuo Tang. The present study was conducted to investigate the pharmacokinetics of furanocoumarin compounds from RAD extract and potential herb-drug interaction in association with metabolism enzymes and transporters. The study revealed that imperatorin (IM) and isoimperatorin (ISOIM), two major bioactive components of natural furanocoumarin had high plasma concentrations and were absorbed and eliminated rapidly with high exposure in liver.
