Type 2 diabetes is the most common metabolic disease in the world affecting more than 250 million people,and is characterized by insulin resistance.Increasing evidence indicates the role of obesity and inflammation in the pathogenesis of insulin resistance and diabetes.We have recently demonstrated that pro-inflammatory cytokine,IL-6 causes insulin resistance,but anti-inflammatory cytokine,IL-10 increases insulin sensitivity.In this talk,I will present our recent findings from transgenic mice with muscle-specific overexpression of IL-10 that are protected from diet-induced insulin resistance in skeletal muscle.Our results indicate that diet-induced obesity increases local macrophages and cytokines in muscle,and this inflammatory response is attenuated in IL-10 expressing mice.Additionally,I will present our recent discovery that inflammation and IL-6 affect cardiac energy metabolism and play an important role in diabetic heart failure,which is a major cause of mortality in diabetes.Overall,my talk will introduce the role of inflammation in insulin resistance,diabetes,and cardiovascular complications.
