  Endocytosis of tyrosine receptor kinases (Trks) is involved in neurotrophin-dependent neuronal survival.However,the mechanisms governing the endocytosis of TrkB and its specific contribution to TrkB downstream signaling have not been explored.In this study,we found that blocking clathrin-and dynamin-dependent endocytosis in neurons of the central nervous system inhibited BDNF-induced activation of Akt but not Erk1/2,while the same blockade in PC 12 cells inhibited NGF-induced activation of Erk 1/2 but not Akt.Moreover,clathrin-and dynamin-dependent endocytosis was required for BDNF-induced neuronal protection and dendritic growth,but not the up-regulation of NPY and ARC.Thus clathrin-and dynamin-dependent endocytosis of Trk receptors may be responsible for differential activation of specific downstream pathways upon neurotrophin binding,leading to regulation of specific neuronal functions.
