The specific binding of proteins to discrete DNA sequences is a fundamental interaction controlling many biological functions.It is important to understand the molecular basis for recognition of DNA,since this will allow both the engineering of proteins to bind at DNA sequences of choice,and the prediction of the DNA recognition sequence among the numerous uncharacterized protein sequences increasingly available from genome sequencing technologies.We have successfully engineered proteins that bind at new,rationally chosen DNA sequences.The bioinformatic approach and experimentation to accomplish this alteration of DNA binding specificity will be described.
