Proteomic Analysis of Vibrio Parahaemolyticus Biofilm at Different Materials

  [Objective]The objective of this study was to determine the preliminary mechanism of differential biofilm formation by Vibrio parahaemolyticus at various food-contact materials.[Methods]Scanning electron microscope was used to compare biofilm architectures at different materials (polystyrene,glass and stainless steel) when cultured at 25 ℃,and meanwhile the proteomic was analyzed through MALDI-TOF-TOF/MS.[Results]The results indicated that V.parahaemolyticus exhibited strongest biofilm-forming ability at glass,polystyrene is next and the last is stainless steel.Meanwhile,the maximum amount of exopolysaccharides and extracelluar protein were obtained at glass.In total,135,123 and 57 differential proteins were detected at glass,polystyrene and stainless steel,respectively.Gene Ontology (GO) analysis indicated that these differentially expressed proteins are related to micromolecule single cell metabolic,cellular localization and biological binding.KEGG pathway analysis showed that the biosynthesis of secondary metabolites pathway plays a more important role on differential biofilm formation on three surfaces.[Conclusion]This study provides basic data for subsequent research of different biofilm formation by V.parahaemolyticus on contact materials,meanwhile provides meaningful goals for further analysis ofbiofilm formation mechanism.
