We have characterised the opioid tetrapeptides endomorphin (EM)-1 and EM-2 in immune tissues of rats and humans.Measured with highly specific radioimmunoassays,endomorphin peptide contents were significantly increased in spleen,thymus and synovial tissues in rats with adjuvant-induced arthritis (AA),a model of chronic inflammation.Synthetic EM-1 injected intraperitoneally significantly attenuated the onset of inflammation in AA.EM-1 and EM-2 were detected by reversed-phase HPLC in intact,undegraded forms within circulating human peripheral blood mononuclear cells.Immunohistochemical staining demonstrated the presence of endomorphins in T cells and macrophages in human synovial tissues.In an ex vivo superfusion system,EM-1 and EM-2 potently inhibited the release of pro-inflammatory cytokines intedeukin (IL)-6 and IL-8 from synovial tissues from patients with osteo-or rheumatoid arthritis.These results demonstrate that endomorphins are endogenously synthesised within immune cells and have the potential to act as potent therapeutic agents in the treatment of chronic inflammatory disease.
