研究目的:血小板衍生生长因子(Platelet-derived growthfactor,PDGF)是人体内一种重要的生长因子.它的受体(PDGF receptor,PDGFR)是酪氨酸激酶家族成员之一.我们前期的研究已经证实了PDGF/PDGFR通路在巨核细胞/血小板生成过程中有重要作用.本项目旨在研究PDGF/PDGFR于原发性血小板增多症发病中的作用,并探索针对其机制的治疗方法.研究方法:用ELISA技术检测原发性血小板增多症患者(n=16)和正常人(n =8)骨髓血清中PDGF-BB的表达;用细胞流式和western blot技术检测PDGF-BB对巨核细胞株JAK2/STAT3、PI3K/AKT信号分子磷酸化水平的影响,及加入酪氨酸激酶抑制剂后,对其作用的影响.
