应用血管生物反应器培养兔颈总动脉血管,通过扫描电镜、HE染色方法考察体外应力培养前、后的兔颈总动脉内皮细胞形态变化。扫描电镜观察显示:应力培养前后内皮细胞均为梭形束状,但培养后内皮细胞短轴间隙加大。HE染色切片表明,培养前血管内皮细胞层清晰可见,培养3 d后血管内皮细胞层连续、相对完整。实验结果表明,利用生物反应器体外应力培养的兔血管内皮细胞基本能够保持形态的完整性,体外应力培养可以作为一种评估手段用于心血管支架材料与血管相互作用在器官水平上的研究。
Rabbit common carotid arteries were cultured with vascular bioreactor. The morphological changes of rabbit common carotid artery endothelial cells were observed by scanning electron microscopy and HE staining before and after stress culture. Scanning electron microscopy showed that the endothelial cells were spindle-shaped before and after stress culture, but the short-axis gap of endothelial cells increased after culture. HE stained sections showed that the vascular endothelial cell layer before culture was clearly visible and the vascular endothelial cell layer was continuous and relatively intact after 3 days of culture. The experimental results show that the rabbit vascular endothelial cells cultured in vitro by bioreactor can basically maintain the integrity of the morphology, and in vitro stress culture can be used as an evaluation method for the study of organ-level interaction of cardiovascular scaffolds and blood vessels.
