Plant oil intake lower ω-6/ω-3 PUFAs ratio in the heart and improve the ischemia/reperfusion injury

  Objective To investigate how does the addition of plant oil in the diet influence the ω-6 /ω-3 PUFAs ratio in the heart and the effect of those on the expression of cytochrome P450 2J (CYP2J) and the myocardial injury due to ischemia/reperfusion.Methods 80 C57BL/6 mice, randomly divided into ordinary diet group and plant oil group.One month later, myocardial ischemia/reperfusion model was built in these mice, 2,3,5 triphenyltetrazolium chloride (TTC) staining was used to measure infarction size.real time reverse transcription-polymerase chain reaction(RT-PCR) and western blot was used respectively to detect the mRNA and protein expression of CYP2J.ω-6/ω-3 PUFAs and EET were tested by high-performance gas chromatography.Results n-6/n-3 PUFA ratio in the heart decreased in the plant oil group than in the ordinary diet group, after ischemia/reperfusion, the expression of CYP2J and the amount of EET in the area at risk was higher in the plant oil group, Infarction size of plant oil group were diminished.Conclusion Properly increase plant oil intake lower the n-6/n-3 PUFA ratio, improve the expression of CYP2J and the ischemia/reperfusion injury.
