Inhibiting the Calcium-binding Protein S100B Restores P53-dependent Apoptosis

  A signature prognostic marker for melanoma is the over expression of a calcium binding protein,S100B,which binds p53 and significantly lowers protein levels of the tumor suppressor.In a human strain of malignant melanoma (C8146A; wild-type p53),we demonstrate here that inhibiting S 100B protein production via siRNA (siRNAS100B) also restores functional p53 protein,phosphorylated p53,and its associated activities including apoptosis (i.e.DNA ladder formation,activation of caspase 3,PARP cleavage).
