硒镉互作对体外培养鸡淋巴细胞中硒蛋白mRNA表达的影响

  镉(Cd)是一种重金属,其毒性对动物的健康构成危险。而硒(Se)是一种重要的营养微量元素。有相关报道指出,镉与硒具有相互的颉颃作用,而二者在禽类中是否存在颉颃作用还未见报道。该研究的目的是通过检测镉和硒对鸡脾脏淋巴细胞中4个内质网相关硒蛋白mRNA的变化,探讨镉毒性和硒保护作用的可能机制。在体外培养鸡脾脏淋巴细胞的培养液中分别添加硒10-7 mol/L、镉10-6 mol/L及硒镉混合物10-7 mol/L和10-6 mol/L,分别在培养的12、24、36和48 h收集细胞用于后续试验的检测。采用实时定量PCR法检测4个内质网硒蛋白,即硒蛋白K、N、S、T的mRNA表达量,结果表明,镉能够降低体外培养鸡淋巴细胞硒蛋白的表达量,而硒能够显著提高硒蛋白mRNA的表达量。通过对硒蛋白之间的相关性分析,表明硒对镉诱导的淋巴细胞损伤具有保护作用。
