Protease-activated receptor 4 (PAR4) is localized in primary sensory neurons and is believed to play an important role in nociceptive response through TRPV1 sensitization.In this study, we examined the effects of PAR4 activation on the TRPVl mRNA and protein expression in cultured rat dorsal root ganglion (DRG) neurons using immunofluorescence,real-time PCR and Western blots after treatment of DRG cultures with AYPGKF-NH2 (PAR4 activating peptide) with or without pre-addition of forskolin (PKA activator) or PKI14-22 (PKA inhibitor).
