Introduction Plateletconcentrationrateinplasma-basedPRPisreportedtobelowerthaninbuffycoat-basedPRPaccordingtopreparation methodsofPRP. Thehypothesisisthatthelowplateletconcentrationplasma-basedPRP(PRPLP)isalsorealPRP. So,weanalyzethecomponentsofthecell,growthfactors,thesecretionmechanicsofthemusingtheautologousconditioned plasma-basedPRP(PRPLP). Materials and Methods WemadePRPLPby49bloodsamples.Each1x106/ulof9samplesofPRPLPhadbeencultured20daysafterhandledbyhuman diplodfibroblasts(HDFs).Duringtheculture,weharvestedmediasupernatants11timesatculturetime30min,1hour,1,2,4,6,9, 10,15,20days.Weanalyzedthequantityofthegrowthfactorsandcytokinesandsecretionmechanics. WecomparedthedataofPRPLP,andwholeblood.ThePRPmadebykitwhichusedsinglespinprocess.Atbloodcomponents, wemeasuredthequantityofplateletandwhitebloodcell.Atgrowthfactors,wemeasuredthequantityofgrowthfactorsand cytokines(IL1-B,PDGF-AA,PDGF-ABBB,VEGF,MMP-13,TGF-B1,TGF-B2,TGF-B3,IGF-1,IGFBP-1,IL-18,andTNF-A)andthe secretionmechanics. Results TheconcentrateofplateletofPRPLPishigherthanthewholeblood.TheconcentrateofwhitebloodcellofPRPLPisthelower thanthewholeblood. Also,concentratesoftheIL-1B,TNF-AwhichareinflammatorycytokinesofPRPLPgroupsislowerthanthethewholeblood. Regardinggrowthfactorsecretionmechanics,averageamountofPDGF-ABBB,IGF-1ishigherthanthethewholeblood AverageamountofcytokineTNF-AandIL-islowerthanthewholeblood(p>.05). PeaktimeofIGF-1fromPRPLPwasshowedearlierthanthewholeblood. PDGF-ABBBwasinitiatorroletoreleaseothergrowthfactorandcytokine. Conclusion ConcentrationofplateletandgrowthfactorswashigherinPRPLPgroupthanwholeblood.But,concentrationofWBCwasvery lowinPRPLPgroup.Especially,secretionmechanicsofgrowthfactorsandcytokinesinPRPLPdifferedfromwholebloods. Inconclusion,eventhoughautologusconditionedplasma-basedPRP(PRPLP)makeslowconcentrationofplateletbutitmightbehaving differentmechanismsoftissuehealing.
