Objective: To investigate the effects of H2S on TGF-β1-induced EMT in human bronchial epithelial cell.Methods: TGF-β1 was applied to induce EMT in 16HBE.H2Swas pre-incubated for 1h and then followed by TGF-β1 for 72 h.EMT markers were examined by western blot.Changes in cell morphology were assessed under phase-contrast light microscopy.Wound healing assay were used to evaluate motility of cells.Results: The cells displayed an elongated shape, with less contact with each other and increased motility after treatment with TGF-β1.In contrast, H2S treatment reversed the cell abnormal morphology and motility induced by TGF-β 1.Moreover, TGF-β1 treatment significantly increased the expression of α-SMA and decreased the expression of E-cadherin.Compared with TGF-β1 group,H2S could reverse these markers by reduced the expression of α-SMA by 45.1% and increase the expression of E-cadherin by 57.6% (P<0.05).Conclusion: Exogenous H2S inhibits TGF-β1-induced EMT in 16HBE.
