目的:观察WY胶囊对慢性轻度不可预见性应激(CUMS)大鼠行为学以及动物脑组织中单胺递质的影响,并探讨WY胶囊对CUMS大鼠抗抑郁作用及其机制.
方法:雄性大鼠72只,实验前对每只动物进行敞箱实验和液体消耗实验,敞箱实验水平运动和垂直运动总分低于30高于120的动物予以剔除,剩余60只动物根据敞箱实验和液体消耗实验的结果把动物随机分为6组,即正常对照组、模型对照组、百优解,WY(1.6、3.2、6.4g原生药/kg)组,每组10只.采用9种刺激随机安排,连续应激大鼠21 d,复制CUMS大鼠抑郁模型.应激当日开始灌胃给药,每日一次,连续2ld.试验期间观察大鼠敞箱运动行为学指标,记录动物体重增长值和糖水消耗百分比.实验结束,采用荧光分光光度法测定每只动物下丘脑和海马组织内去甲肾上腺素(NE),5-羟色胺(5-HT),多巴胺(DA),5-羟吲哚乙酸(5-HIAA)的含量.
结果:①WY胶囊高、中、低剂量组动物体重增长值均显著高于模型组.②WY高、中剂量水平运动格数明显高于模型对照组.③WY胶囊高、中、低组糖水百分比均显著高于模型组.④WY高剂量组海马组织中NE、DA含量明显升高,下丘脑组织单胺递质NE、DA、5-HT含量明显升高.
结论:WY胶囊对CUMS大鼠体重降低、自发活动减少及大鼠蔗糖饮用水量下降等行为有改善作用,其机制可能是通过影响动物海马及下丘脑组织中NE、DA、5-HT等单胺递质含量变化而发挥作用.
