New evidence of TCDD toxicity metabolomics for the old environmental toxicant

2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) has been listed as the most toxic organic pollutant.However,report on its poisoning case was rare.In this study,toxic effect of TCDD in blood,liver and skeletal muscle tissues of C57BL/6J (C6) and DBA/2J (D2) mice was investigated by using LC-QTOFMS-based metabolomics.With the non-targeted approach in conjunction with multivariate data analysis,hundreds of ions were aligned and extracted with P-value and fold-change in mice exposed to TCDD compared to control group.As a result,in the aryl hydrocarbon receptor (AhR)-sensitive mouse model-C6,fatty acid beta-oxidation was markedly enhanced.Meanwhile,perturbation of other metabolic pathways was also observed,namely the accumulation of fatty acids and lysophospholipids,the reduced amino acid biosynthesis and the decreased purine metabolism.In contract,in the less-sensitive mouse model-D2.
