我国非人灵长类十二指肠贾第虫多位点分析及地理隔离和宿主适应性集聚体B的发现

  为调查和研究贾第虫的流行性和基因型,使用SSU rRNA,tpi,gdh,和bg基因多位点分析方法,对采集的43个种类1,882份非人灵长类的粪便样品进行检测盒分析.结果显示,基于tpi基因,22个种类的122份(6.5%)非人灵长类的样本检测到贾第虫,包括恒河猴(2.7%),阿萨姆邦猕猴(11.1%),食蟹猴(10.0%),长尾猕猴(52.9%),日本猕猴(33.3%),白头叶猴(0.7%),赤猴(31.3%),白须猴(100%),叶猴(9.1%),金丝猴(13.2%),黑白疣猴(14.3%),白腹长尾猴(16.7%),松鼠猴(22.2%),绿猴(17.7%),懒猴(50.0%),环尾狐猴(56.5%),山魈(5.1%),阿拉伯狒狒(3.7%),黄狒狒(40.0%),橄榄狒狒(11.8%),白颊长臂猿(14.3%),婆罗洲猩猩(19.2%).对于不同的地区,郑州动物园的感染率为9.1%,石家庄动物园11.2%,武汉动物园7.6%,太原动物园13.6%,长沙野生动物园44.0%,北京动物园22.2%,上海动物园8.2%,上海野生动物园20.9%,以及河南,广西和广东猴场的感染率为2.9%.检测到的贾第虫为集聚体A(n=4)和集聚体B(n=118).对tpi基因序列的分析显示,集聚体A包括A1,A2和一个新的亚型.对tpi,gdh和bg基因的多位点分析共检测到18(7个已知和11个未报到),11(5已知和6个未报到)和9(2已知和7个未报到)的亚型在115,65和35个集聚体B分离株中.所有3个基因位点均成功扩增的32个集聚体B分离株中,共发现15个多位点基因亚型(MLGS),包括2已知和13未报到MLGs.集聚体B的串联序列的系统进化发育分析显示,这里发现的MLGs与来自人类,非人灵长类,兔和意大利和瑞典的豚鼠具有遗传差异.也进一步表明,环尾狐猴和松鼠猴分离的集聚体B与其他非人灵长类的可能存在差异.
