Expression of Ano1, a newly-found calcium-activated chloride channel,in rat hearts

  The calcium-activated chloride channels (CaCCs) are critical for many physiolo-gical functions.In cardiac tissues, CaCCs play important roles in the regulation of cardiac excitability.Although Anol (or TMEM16A) has been identified as CaCCs in many tissues, the molecular identity of CaCCs in heart remains unknown.In this study, we examined the expression of Anol in rat heart by methods of reverse transcription polymerase chain reaction (RT-PCR) and Western blotting.RT-PCR shows the existence of Anol mRNA in heart tissue;while Anol protein expression were further confirmed by Western blotting, using HEK293 cells transfected with Ano1 cDNA as a positive control.The expression of heart Ano1 protein is higher in neonatal rats than that in adults.Furthermore, we noticed that phosphate-dylinositol 3-kinase (PI3K) inhibitor (LY294002) could up-regulate Ano1 expression in primarily cultured neonatal rats cardiomyocytes.Whole-cell patch-clamp studies showed that primarily cultured neonatal rat cardiomyocytes expressed CaCC currents.Conclusion: Our results strongly suggest that Ano1 protein exists in rat cardiomyocytes, and Ano1 expression may be regulated by PI3K pathway.
