Chromosome 17 is frequently abnormal in breast cancer.Several genes important to breast cancer treatment and prognosis reside on this chromosome, including HER2, topoisomerase Ⅱ α and P53.In previous studies, we analyzed breast cancers using bacterial artificial chromosome arrays and showed that results of HER2 analysis by comparative genomic hybridization arrays are accurate and robust in a clinical validation study (Yeh et al, Modem Pathology 2009,May 15 Epub).With the more recent availability of oligorarrays with increased probe density, we can further analyze chromosome 17 in breast cancers with greater precision.In this study, 100 frozen samples of invasive lobular and invasive ductal carcinoma, with known immunohistochemistry and fluorescence in situ hybridization results for HER2,were analyzed by comparative genomic hybridiz.ation to a commercially available oligoarray.We demonstrate the results of analysis of chromosome 17, with attention to the HER2 amplicon and the centromere region, as well as other genes of interest, such as topoisomerase Ⅱ α.
