Myostatin调控表达的miR-A在肌生成过程中发挥重要功能

  已有数据显示Myostatin负调控肌肉干细胞的激活及自我更新过程,然而其分子机制并不明确。近年来的一些研究表明microRNAs在该过程中发挥重要作用。因此microRNAs可能参与Myostatin调控的干细胞功能。我们分别从Myostatin敲除小鼠及野生型小鼠中提取骨骼肌组织用并用微阵列分析法预测到受Myostatin负调控的miR-A,并且在Myostatin处理的C2C12细胞中也得到了证实。体内研究发现,与野生型小鼠相比在发育过程中miR-A转基因小鼠的干细胞数目及Pax7表达量均较低。同时我们在CTX肌肉损伤再生模型中发现Pax7及myoD表达量在miR-A转基因小鼠中均高于野生型小鼠,这些数据提示着miR-A可能在骨骼肌干细胞的激活及自我更新过程中有重要作用。结论:我们的数据显示miR-A受Myostatin调控并在骨骼肌干细胞的激活及自我更新过程中有着至关重要的功能。
