Objective: To investigate a new adoptive immunotherapy method for renal cell carcinoma.Methods: Human umbilical cord blood (UCB) was obtained from volunteer mothers at the Affiliated Hospital of GuangdongMedical College.UCB mononuclear cells were prepared by centrifugation.The cells were co-cultured on a layer of precultured stromal cells overnight.Mouse stromal cell line OP9was used for the co-culture.After one or two weeks, floating cells were transferred to a new layer of stromal cells and incubated for a month or more to observe the emergence of proliferative blastlike cells.These cells were called mouse stromal cell-activated cord blood monocyte (MSCACBM).Lymphokine-activated cord blood monocytes (LACBMs), which were induced by recombinant IL-2, and the cord blood monocyte (CBM) were used as control samples.The cell morphology was observed via phase-contrast microscopy.Surface molecules, such as CD4,CD8 for subtype lymphocyte, and CD57 for NK cells, were determined via flow cytometry.The cytotoxic activity of these three kinds of effector cells on renal cell carcinoma OS-RC-2cells was measured via MTT assay.
