六价铬诱导干细胞DNA损伤的机理研究

  引言:干细胞是一类具有自我更新和分化潜能的细胞,是个体生长发育的基础。工业科技高度发达的今天,各种环境污染无时无刻不威胁着人类的健康。诸多的毒性物质容易造成DNA的损伤而诱发疾病。六价铬Cr(Ⅵ)是一种环境致癌物,其在诱导多潜能干细胞(induced pluripotent stem cells,iPSCs)和精原干细胞(spermatogonial stem cells,SSCs)中的基因组毒性及其机制尚未见报道。鉴于多潜能干细胞iPSCs和成体干细胞SSCs在人类疾病模型和再生医学研究中的潜在价值,了解这些细胞在经历环境有毒物质如何保护基因组稳定性是非常重要的。本研究关于六价铬诱导干细胞DNA损伤调节机制的研究发现,可以为预防和治疗环境污染导致的人类疾病,以及更好的应用于再生医学和药物研发提供科学资料。
