Histaminergic output from the tuberomammillary nucleus (TMN) is considered to play a crucial role in mediating arousal.Under physiological conditions,the extracellular histamine levels in the preoptic/anterior hypothalamus areas of cats are the highest during waking,lower during non-rapid eye movement (non-REM) sleep,and the lowest during REM sleep.On the other hand,enhancement of histaminergic neuronal activity by either orexin or EP4 agonist produces arousal.Conversely,sleep is promoted by decreasing histaminergic transmission through pharmacological blockade of central histaminergic receptors,inhibition of histidine decarboxylase (HDC),a key enzyme for histamine biosynthesis,and hyperpolarization of the TMN with GABAergic agonists.Adenosine is an endogenous sleep-inducing substance.
