Objective: Only two splice orexin (OX) variants (OXA and OXB) derived from a unique precursor have been identified.We test the different effects of OXA and OXB on breathing in the Pre-Botzinger Complex (PBC).Methods: Different concentrations of OXA or OXB were microinjected into the PBC to observe the change of phrenic nerve discharge integral (PNDI), blood pressure (BP), heart rate (HR) and respiratory frequency (RF) in normal SD rats.Result: The maximum PNDI ratio in response to microinjection of 1 μM and 5 μM OXA was 3.12±0.36 and 2.93±0.41 compared with a value of 1.00±0.00 following injection of saline (n=7, P<0.01), that of 1 μM and 5 μM OXB was 2.16±0.44 and 1.48±0.37 (n=7, P<0.05), while BP, HR and RF did not show significant differences.Conclusion: The results suggested that both OXA and OXB enhance respiratory activity in the PBC,the effect of OXA is seemed stronger then that of OXB.
