1.ER-positive breast cancer.We are conducting a systematic investigation of cross-talk between Notch and other pathways in breast cancer,with the objective of designing rational therapeutic combinations for pre-clinical and clinical investigation.We have delineated a bi-directional cross-talk between the estrogen receptor alpha (ERα) and Notch.Estrogen inhibits Notch transcriptional activity,possibly by modulating Notch processing by ADAMs.Conversely,Notch-1,but not Notch-2 or Notch-4,stimulate ERα transcriptional activity in the absence of estrogen.
