人α-乳清白蛋白（HLA）是一种小分子量的酸性Ca2+结合蛋白,不仅具有很高的营养价值,而且还具有促进幼儿的神经发育、调节睡眠、刺激人体免疫等多种生物学功能。新近的研究发现,α-乳清白蛋白还具有杀菌和抗癌的作用。为了研究人α-乳清白蛋白在转基因番茄中的表达。首先,我们构建了植物表达载体pBRALA,该载体具有完整的植物表达元件,具有CaMV35启动子、农杆菌T-DNA左右边界、植物报告基因gus和植物选择标记基因hpt,适用于对根癌农杆菌的转化。其次,我们通过冻融法转化,将pBRALA导入到了农杆菌LBA4404中。再次,我们以番茄的子叶和下胚轴为材料,探讨了各种因素对番茄再生的影响,建立了一套适合番茄的再生体系。最后,通过叶盘法将人α-乳清白蛋白基因导入番茄,经筛选,获得抗性番茄植株18株;经PCR扩增和GUS染色检测,初步证明外源基因已经转入到其中的12株中。转基因番茄植株经培养,开花并结果。
