目的探讨邻苯二甲酸二乙基己基酯(DEHP)对大鼠肝细胞和睾丸细胞DNA的损伤。方法选用雄性Wis-ter大鼠25只,随机分为5组,每组5只,通过灌胃的方法摄入DEHP,4个染毒组剂量浓度分别为1500、3000、4500、6000mg·kg-1.d-1,1个阴性溶剂对照组,用单细胞凝胶电泳技术(彗星试验)检测大鼠肝细胞和睾丸细胞DNA的损伤。结果各染毒组和对照组大鼠肝细胞DNA损伤率分别为28.0%、69.0%、86.0%、94.0%和18.0%,各染毒组肝细胞DNA损伤程度与对照组比较差异有显著性(P<0.05,P<0.01),且大鼠肝细胞损伤率与染毒剂量之间呈剂量-反应关系(r=0.942,P<0.01);各染毒组和对照组大鼠睾丸细胞DNA损伤率分别为29.0%、71.0%、89.0%、95.0%和15.0%,各染毒组睾丸细胞DNA损伤程度与对照组比较差异有显著性(P<0.05,P<0.01),且大鼠睾丸细胞损伤率与染毒剂量之间呈剂量-反应关系(r=0.930,P<0.01)。结论DEHP可造成大鼠的肝细胞和睾丸细胞DNA的损伤,并随染毒剂量的增加有加重的趋势。
Objective To investigate the DNA damage of rat liver cells and testicular cells induced by diethylhexyl phthalate (DEHP). Methods 25 male Wis-ter rats were randomly divided into 5 groups with 5 mice in each group. DEHP was ingested by intragastric administration. The dose of 4 groups were 1500, 3000, 4500 and 6000 mg · kg- 1.d-1, 1 negative solvent control group, single-cell gel electrophoresis (comet assay) to detect DNA damage in rat hepatocytes and testicular cells. Results The DNA damage rates of hepatocytes in the treated and control groups were 28.0%, 69.0%, 86.0%, 94.0% and 18.0%, respectively. The DNA damage of hepatocytes in each exposure group was significantly different from that in the control group (P <0.05, P <0.01). The rate of hepatocellular injury in rats was dose-response relationship with the dose (r = 0.942, P <0.01) (P <0.05, P <0.01). The DNA damage of testis cells in each exposure group was significantly different from that in control group (P <0.05, P <0.01) There was a dose-response relationship between the injury rate and exposure dose (r = 0.930, P <0.01). Conclusion DEHP can cause DNA damage in rat hepatocytes and testicular cells, which may aggravate with the dose of DEHP.
