背景:正常成年动物中枢神经系统内存在能增殖、多向分化潜能的神经干细胞或神经前体细胞。在生理情况下,这些内源性神经干细胞或前体细胞处于静息状态。当脑缺血时,这些细胞将处于何种状态?目的:研究大鼠局灶性脑缺血时,内源性神经干细胞的分布、增殖、分化。设计:以大鼠为研究对象,随机对照的探索性研究。单位:一所医学院的神经生物学研究室、组织胚胎学教研室。材料:实验于2001-07/2002-07在泸州医学院神经生物学研究室完成。选择健康成年SD大鼠41只,雌雄不限,体质量250~300g。干预:阻塞大鼠大脑中动脉2h再灌流0.5,3,6,12h,1,2,3,5,10d制成局灶性脑缺血模型,阻塞线不能阻塞大脑中动脉为假手术组,正常大鼠为对照组。在各时间点处死动物,制成脑组织切片,用免疫组织化学单标和双标观察内源性神经干细胞或前体细胞的增殖、分布、分化。主要观察指标:内源性神经干/前体细胞的分布、分化。结果:在正常组和假手术组多数脑室室管膜细胞增殖性细胞核抗原免疫反应阳性,脑实质内有零星散在增殖性细胞核抗原阳性细胞。再灌流3h,增殖性细胞核抗原阳性细胞出现在嘴侧脑室下层。再灌流12h以后,双侧侧脑室的一些脉络膜丛细胞增殖性细胞核抗原免疫反应阳性。再灌流3~10d,视前区梗死区周边、纹状体、额顶皮质的增殖性?
BACKGROUND: Normal adult animals have neural stem cells or neural progenitor cells capable of proliferating and multipotentiating in the central nervous system. Under physiological conditions, these endogenous neural stem cells or precursor cells are at rest. When the brain ischemia, these cells will be in what state? Objective: To study the focal cerebral ischemia in rats, the distribution of endogenous neural stem cells, proliferation and differentiation. Design: A randomized, controlled study of rats. Unit: a medical school neurobiology laboratory, Department of Embryology. MATERIALS: Experiments were performed at the Department of Neurobiology, Luzhou Medical College from July 2001 to July 2002. Choose 41 healthy adult SD rats, male or female, body weight 250 ~ 300g. Intervention: occlusion of rat middle cerebral artery 2h reperfusion 0.5,3,6,12 h, 1, 2, 3, 5, 10d made of focal cerebral ischemia model, obstruction line can not block the middle cerebral artery as sham operation group, Normal rats as control group. Animals were sacrificed at each time point to make brain tissue sections. The proliferation, distribution and differentiation of endogenous neural stem cells or precursor cells were observed by immunohistochemical single and double labeling. MAIN OUTCOME MEASURES: Distribution and differentiation of endogenous neural stem / precursor cells. Results: In the normal group and the sham operation group, most of the ventricular ependymal cells were positive for proliferating cell nuclear antigen immunopositive. There were sporadic scattered proliferating cell nuclear antigen positive cells in the brain parenchyma. Reperfusion 3h, proliferating cell nuclear antigen positive cells appear in the lower part of the mouth of the ventricle. After 12 h of reperfusion, some choroidal plexus proliferating cell nuclear antigen immunoreactivity was positive in bilateral lateral ventricle.
