目的探讨结缔组织生长因子(CTGF)在子宫颈癌组织中的表达水平及其与临床病理因素间的关系。方法用实时荧光定量聚合链式反应和免疫印迹法检测61例子宫颈癌癌组织(试验组)和对应癌旁正常宫颈组织(对照组)中CTGF mRNA及蛋白的表达水平,并进行多因素Logistic回归分析。结果对照组和试验组的CTGF mRNA表达量分别为(1.00±0.01)和(0.30±0.03),CTGF蛋白的表达量分别为(0.98±0.02)和(0.22±0.05),差异均有统计学意义(均P<0.01)。多因素Logistic回归分析结果显示:CTGF mRNA表达与宫颈癌的FIGO分期、淋巴结转移和分化程度呈负相关,CTGF蛋白表达与FIGO分期呈负相关。结论 CTGF在宫颈癌中低表达,可能作为抑癌因子在宫颈癌的发生发展中发挥作用。
Objective To investigate the expression of connective tissue growth factor (CTGF) in cervical cancer and its relationship with clinicopathological factors. Methods The expression of CTGF mRNA and protein in 61 cases of cervical cancer (corresponding to experimental group) and corresponding normal cervical tissues (control group) were detected by real-time fluorescence quantitative polymerase chain reaction and Western blotting, and multivariate Logistic regression analysis. Results The expression of CTGF mRNA in the control group and the experimental group were (1.00 ± 0.01) and (0.30 ± 0.03), respectively, and the CTGF protein expression was (0.98 ± 0.02) and (0.22 ± 0.05), respectively (All P <0.01). Multivariate Logistic regression analysis showed that CTGF mRNA expression was negatively correlated with FIGO staging, lymph node metastasis and differentiation of cervical cancer, and CTGF protein expression was negatively correlated with FIGO staging. Conclusion CTGF is low expressed in cervical cancer and may play a role as a tumor suppressor in the development of cervical cancer.
